First, synthesis line of 5-Chlorothiophene-2-carboxylic Acid CAS NO. 24065-33-6 as follows
Step 1: Preparation of 5-Chlorothiophene-2-aldehyde
Add 200 grams of N, N-dimethylformamide (DMF) and 118.5 grams (1.0 mole) of 2-chlorothiophene to a 1-liter dry four necked bottle equipped with stirring, thermometer, condenser, and constant pressure dropping funnel. Heat and maintain the internal temperature between 40-60 ℃. Slowly drop 169 grams (1.1 mole) of phosphorus oxychloride from the constant pressure dropping funnel. After dropping, stir at 95-100 ℃ for 2 hours. Cool to 20 ℃, pour the reaction solution into a beaker containing 400 grams of crushed ice and 400 grams of water, then neutralize with 30% sodium hydroxide solution to a pH of 6-7, separate the oil layer, extract the water layer with dichloromethane three times, each time using 300 grams of dichloromethane, combine the dichloromethane layer and the separated oil layer, wash the organic phase with 50 grams of water once, distill the organic phase to recover dichloromethane, and then vacuum distill to obtain 141.5 grams of 5-chlorothiophene-2-aldehyde, with a yield of 96.6% and a GC purity of 99.97%.
Step 2: Preparation of 2-chlorothiophene-5-carboxylic acid
Add 200 grams of dichloromethane, 29.5 grams (0.2 moles) of 5-chlorothiophene-2-aldehyde, 30 grams of 30% hydrogen peroxide, and stir at 25-30 degrees Celsius for 6 hours to a 500 milliliter four necked bottle with stirring and a thermometer. Layered, the aqueous layer was extracted three times with 50 grams of dichloromethane each time. The dichloromethane layers were combined, and the organic phase was washed once with 20 grams of a 5% sodium sulfite aqueous solution. The organic phase was distilled to recover dichloromethane, and the residue was recrystallized from petroleum ether to obtain 30.5 grams of white solid 2-chlorothiophene-5-carboxylic acid with a yield of 93.8% and HPLC purity of 99.98%.

The method for preparing 2-chlorothiophene-2-carboxylic acid chloride using 2-chlorothiophene-5-carboxylic acid is as follows:
Add 100 grams of dichloromethane, 16.5 grams (0.1 mole) of 2-chlorothiophene-5-carboxylic acid, 1 drop of N, N-dimethylformamide, 15 grams of thionyl chloride, and stir at 40 degrees Celsius for 4 hours to a dry 250 milliliter four necked bottle connected to a stirring, thermometer, and alkali absorption system. Distillation was used to recover dichloromethane and excess thionyl chloride, followed by vacuum distillation (80-90 ℃/5 mmHg) to obtain 18.0 grams of colorless liquid 5-chlorothieno-2-formyl chloride with a yield of 100.0% and a GC purity of 99.98%.
